چکیده

Background Despite the important role of amyloid-beta protein (Aβ) in initiation and progression Alzheimer’s Disease (AD), precise mechanism by which Aβ disrupts neuronal activity (NA) structure during disease development, whether this is an appropriate therapeutic target still controversial. Recently, we introduced several novel NA metrics (Kazemeifar, PLoS One 2017; Haddad, ISMRM 2019) based on specific spatiotemporal features resting-state fMRI (rs-fMRI) signal. These are significantly lower AD mild cognitive impairment (MCI) compared to normal elderly controls. Here, examine there association between levels deposition grey matter (GM) measured PET MCI. Method [18F]florbetapir images rs-fMRI data (TR = 3s, 197 volumes) 9 participants from Neuroimaging Initiative (ADNI, 3 early MCI (EMCI), MCI, AD, aged 77 ± 7 years, females) were included. The was pre-processed decomposed into independent components (ICs) using IC analysis. ICs associated with identified a support vector machine classifier (Demertzi, Cortex 2014). quantified each voxel magnitude This calculated similarity (cross-covariance) signal (Haddad, 2019). In subject, (Jagust, Alzheimers. Dement. 2015) registered T1-weighted image maps already registered. GM voxelwise correlation then conducted uptake subject. Result one EMCI subject (Figure 1) related scatter plot 2) show GM. all subjects studied, negatively correlated (r -0.25 0.14) (Table 1). Conclusion negative preliminary study indicates that contributes decreased rs-fMRI. Future studies should determine susceptibility various brain regions accumulation.

Identification of mild cognitive impairment disease using brain functional connectivity and graph analysis in fMRI data

Background: Early diagnosis of patients in the early stages of Alzheimer's, known as mild cognitive impairment, is of great importance in the treatment of this disease. If a patient can be diagnosed at this stage, it is possible to treat or delay Alzheimer's disease. Resting-state functional magnetic resonance imaging (fMRI) is very common in the process of diagnosing Alzheimer's disease. In th...

Dissociation between Brain Amyloid Deposition and Metabolism in Early Mild Cognitive Impairment

BACKGROUND The hypothetical model of dynamic biomarkers for Alzheimer's disease (AD) describes high amyloid deposition and hypometabolism at the mild cognitive impairment (MCI) stage. However, it remains unknown whether brain amyloidosis and hypometabolism follow the same trajectories in MCI individuals. We used the concept of early MCI (EMCI) and late MCI (LMCI) as defined by the Alzheimer's d...

Diagnosed Mild Cognitive Impairment Due to Alzheimer’s Disease with PET Biomarkers of Beta Amyloid and Neuronal Dysfunction

The aim of this study is to identify mild cognitive impairment (MCI) due to Alzheimer's disease (AD) using amyloid imaging of beta amyloid (Aβ) deposition and FDG imaging of reflecting neuronal dysfunction as PET biomarkers. Sixty-eight MCI patients underwent cognitive testing, [11C]-PIB PET and [18F]-FDG PET at baseline and follow-up. Regions of interest were defined on co-registered MRI. PIB ...

Different Characteristics of Cognitive Impairment in Elderly Schizophrenia and Alzheimers Disease in the Mild Cognitive Impairment Stage

We compared indices of the revised version of the Wechsler Memory Scale (WMS-R) and scaled scores of the five subtests of the revised version of the Wechsler Adult Intelligence Scale (WAISR) in 30 elderly schizophrenia (ES) patients and 25 Alzheimer’s disease (AD) patients in the amnestic mild cognitive impairment (aMCI) stage (AD-aMCI). In the WMS-R, attention/concentration was rated lower and...

Monitoring disease progression in mild cognitive impairment: Associations between atrophy patterns, cognition, APOE and amyloid

BACKGROUND A disease severity index (SI) for Alzheimer's disease (AD) has been proposed that summarizes MRI-derived structural measures into a single score using multivariate data analysis. OBJECTIVES To longitudinally evaluate the use of the SI to monitor disease progression and predict future progression to AD in mild cognitive impairment (MCI). Further, to investigate the association betwe...
